Pivotal Trial

This is an open-label extension study designed to evaluate the long-term safety and tolerability of continued setmelanotide treatment in male and female patients ≥2 years of age who have completed or transitioned from a previous study with setmelanotide for rare genetic, syndromic, or acquired diseases of obesity upstream of the MC4R pathway.

I have experienced new or worsening depression with suicidal thoughts or behaviors.

My family (parents or siblings) or I have a history of melanoma.

I am over 2 years old and have previously participated in a setmelanotide study.

I have a condition treatable with setmelanotide (IMCIVREE®).

The current study assesses the tolerability and efficacy of combination therapy with PD-1 (nivolumab) and pan-RAF-kinase (Tovorafenib) inhibition for the treatment of children and young adults with craniopharyngioma.

I have not received any treatment for my cancer yet.

I do not have unmanaged symptoms from neuroendocrine issues.

I still have visible signs of my disease while on maintenance therapy.

I haven't had chemotherapy or radiotherapy recently.

I do not need urgent surgery or radiation due to rapidly worsening symptoms.

I am not currently ill with a condition that is not under control, nor am I participating in another clinical trial.

I am not currently taking any medications that would exclude me from the trial.

I have been newly diagnosed with craniopharyngioma based on imaging.

I am eligible for surgery to remove or sample a piece of my tumor.

I can attend all required follow-up visits.

I do not have ongoing severe nausea, problems absorbing food, or major bowel/stomach surgery.

I have not had pneumonitis or chest radiation in the last 5 years.

My craniopharyngioma has come back and is confirmed by a biopsy.

I can do most activities but may need help.

I do not have active heart disease or a recent heart attack.

MEKTOVI (binimetinib) is an oral, highly selective reversible inhibitor of mitogen-activated extracellular signal regulated kinase 1 (MEK1) and MEK2. The biological activity of binimetinib that has been evaluated bith in vitro and in vivo in a wide variety of tumor types In this Phase II, the drug will be used to treat pediatric patients diagnosed with recurrent Adamantinomatous Craniopharyngioma including patients who have undergone surgery and/or radiation therapy.

MEKTOVI for Pediatric Craniopharyngioma

This trial tests MEKTOVI, a pill that targets cancer growth proteins, in children and young adults with a recurring brain tumor called Adamantinomatous Craniopharyngioma. The drug works by blocking signals that make cancer cells grow.

I do not have any major health or surgical issues that could affect my safety or participation in the study.

I have ACP, had surgery, but no prior radiation. I may have had other treatments.

I haven't had live vaccines recently and don't have an uncontrolled infection.

I have not had recent surgery, substance abuse issues, or specific infections.

I have waited the required time after my last therapy dose.

My heart, kidneys, liver, brain, and bone marrow are working well.

I am mostly able to care for myself, regardless of my age.

I have severe stomach issues or can't take pills properly.

I haven't been on a stable or reducing dose of steroids for the last week.

My ACP has worsened or come back 6 months after finishing radiation.

I have recovered from the side effects of my previous cancer treatments.

I have been diagnosed with adamantinomatous craniopharyngioma.
