What is Supprelin

HistrelinThe Generic name of this drug
Treatment SummaryHistrelin is a hormone-based medicine used to treat a variety of conditions. It is delivered in an implant that slowly releases medicine into the body to inhibit the release of hormones from the pituitary gland. It is approved by the FDA as Supprelin LA for the treatment of central precocious puberty in children and as Vantas for palliative treatment of advanced prostate cancer in adults.

Effectiveness

How Supprelin Affects PatientsHistrelin is a hormone-regulating drug used to treat central precocious puberty (CPP) in children. It works by blocking the hormone-producing receptors in the pituitary gland, which in turn reduces the production of luteinizing and follicle-stimulating hormones. This slows down the growth velocity, allowing the child to potentially reach their predicted adult height. Histrelin also causes a temporary increase in sex hormone production in the body. When using Histrelin, laboratory tests should be conducted to monitor hormone levels. For CPP, this includes LH, follicle-stimulating hormone and est
How Supprelin works in the bodyHistrelin is a drug that blocks the body's hormones responsible for sexual maturation and reproductive function. When used as an implant, it delivers a continuous dose of the drug. Histrelin acts as a GnRH agonist, which binds to the GnRH receptor and activates it. This causes an increase in circulating levels of LH and FSH hormones. However, over time, the continuous administration of histrelin desensitizes the GnRH receptor and lowers LH and FSH levels. In children with central precocious puberty, this lowers the level of sex steroids and reduces height potential. In adults with prostate cancer, it lowers

Mobocertinib
Major
The risk or severity of QTc prolongation can be increased when Histrelin is combined with Mobocertinib.
Ziprasidone
Major
The risk or severity of QTc prolongation can be increased when Histrelin is combined with Ziprasidone.
Supprelin Toxicity & Overdose RiskTests on animals have shown that histrelin acetate is safe up to 200 mcg/kg for rats and rabbits, and 2000 mcg/kg for mice. This is much higher than the recommended human dose of 10 mcg/kg/day. Studies of patients who received one, two, or four implants of histrelin had similar side effects. There have been no reported cases of overdose in clinical trials of histrelin products, and accidental or intentional overdose is unlikely due to the constant dose delivered by the implants.

What does Supprelin LA treat?

"SUPPRELIN LA is a medication that is implanted under the skin and releases gonadotropin-releasing hormone (GnRH). It is used to treat children who have precocious puberty, a condition in which sexual maturity begins too early."

How long does histrelin implant last?

"A single histrelin implant is effective in treating central precocious puberty for 2 years, according to a new study. The study was funded by the National Institutes of Health and was published in the New England Journal of Medicine. histrelin is a synthetic hormone that is similar to gonadotropin-releasing hormone.

What does the supprelin implant do?

"An implantable medication, Supprelin LA (histrelin acetate) is used to prevent further progression of puberty. The implant is inserted under the skin on the inside of the upper arm and slowly releases medication into the tissues. This medication is used in cases of precocious puberty or gender dysphoria."
